Evaluation of [18F]AlF-EMP-105 for Molecular Imaging of C-Met

C-Met is a receptor tyrosine kinase that overexpressed in range of different cancer types, and has been identified as potential biomarker for imaging therapy. Previously, 68Ga-labelled peptide, [68Ga]Ga-EMP-100, shown promise c-Met renal cell carcinoma humans. Herein, we report the synthesis preliminary biological evaluation an [18F]AlF-labelled analogue, [18F]AlF-EMP-105, by positron emission tomography. EMP-105 was radiolabelled using aluminium-[18F]fluoride method with 46 ± 2% RCY >95% RCP 35–40 min. In vitro showed [18F]AlF-EMP-105 high specificity c-Met-expressing cells. Radioactive metabolite analysis at 5 30 min post-injection revealed good blood stability, but undergoes transformation—transchelation, defluorination or demetallation—in liver kidneys. PET non-tumour-bearing mice radioactive accumulation kidneys, bladder urine, demonstrating tracer cleared predominantly [18F]fluoride system. With its expressing cells, shows diagnostic tool cancer.
